Effects of various cryoprotective agents on the survival of unfrozen and frozen mouse embryos.

چکیده
The effects of DMSO, ethylene glycol, glycerol, erythritol, dimethylformamide and sucrose on the survival of unfrozen and frozen mouse morulae were examined. All the agents had a deleterious effect on survival of the unfrozen morulae at 20 degrees C. The harmful effect of erythritol was lower at 0 degrees C and sucrose had a protective effect at that temperature. Dimethylformamide and DMSO were more harmful than ethylene glycol and glycerol at both temperatures. Higher proportions of the morulae frozen in DMSO developed into expanded blastocysts when slowly frozen samples were thawed slowly (82%) than when they were thawed rapidly (85%) than when they were thawed slowly (4%). When the samples were frozen in the presence of ethylene glycol or glycerol, higher survival rates were obtained after slow (80--94%) than rapid (1--59%) freezing. When erythritol was used, only embryos thawed rapidly developed (31--50%) in subsequent cultures. Sucrose and dimethylformamide did not afford cryoprotection. Morulae frozen--thawed rapidly in the presence of ethylene glycol, glycerol or erythritol and transferred to recipients developed into normal young.

In-vitro development of refrozen mouse embryos.
To evaluate the effects of sequential, repetitive freezing on their in-vitro development, mouse embryos at the eight- to 16-cell stage were subjected to one of five treatments. They were (i) cultured as unfrozen controls, (ii) frozen once and cultured, (iii) subjected to two consecutive freeze-thaw cycles, (iv) frozen and thawed, and then cultured for 18-30 h before being frozen a second time, ...
متن کاملEffect of vitrification on viability and chromosome abnormalities in 8-cell mouse embryos at various storage durations.
This study was designed to investigate the effect of vitrification and post-thaw survival and chromosomal aberrations caused by vitrification of vitrified 8-cell mouse embryos in comparison with a control group. To this purpose the survival rate and the frequency of chromosomal aberrations were assessed in frozen-thawed 8-cell mouse embryos after various storage durations in the presence of eth...
متن کاملEffects of sperm pretreatment on efficiency of ICSI-mediated gene transfer in pigs.
Intracytoplasmic sperm injection (ICSI)-mediated gene transfer has recently been shown to be an effective technique for producing transgenic pigs; however, the types of sperm pretreatment having the most beneficial effects on post-ICSI embryogenesis or transgenic efficiency have not been clarified. In the present study, we performed ICSI-mediated gene transfer using pig sperm subjected to vario...

متن کاملP-107: The Effects of Cryotop Vitrification on Heat Shock Protein 72 Expression in Mouse 2-Cell Embryos by Nested Quantitative PCR
Background: The aim of the study was to compare the effects of two different concentrations of cryoprotectants by Cryotop vitrification on survival and Heat shock protein 72 (Hspa1a) expression of two-cell mouse embryos. Materials and Methods: Different cryoprotectants’ concentrations of the combination of dimethyl sulfoxide (DMSO) and ethylene glycol (EG) were used and compared with each other...
